| Principle | Direct magnesium determinations employing spectrophotometric methods without deproteinization of the samples have been described. These use calmagite, methylthymol blue and Titan yellow. The sodium salt of Xylidyl Blue is 4-hydroxy-3-[2-hydroxy-3-(2, 4-dimethyl-phenyl amino carbonyl)-1-naphthylazo] benzene sulfonic acid and is used in our procedure. This compound forms a red complex in alkaline solution with magnesium. The absorbance at 520 nm of the red Xylidyl Blue l: magnesium complex is proportional to the concentration of magnesium in the sample. Interference by calcium is prevented by the use of EGTA. |
